Adding East-Asian LRRK2 variants and other Taiwan-specific loci to a European-derived polygenic risk score improved discrimination from AUC 0.59 to 0.62 in a held-out Taiwanese test set.
PRS-EUR achieved an AUC of 0.590 (95% CI, 0.574–0.607). Adding 11 East Asian lead variants (Foo et al., 2020) yielded PRS-EUR-Asia and improved AUC to 0.606. Further inclusion of Taiwan-specific variants [including LRRK2 p.G2385R, p.R1628P] increased it to 0.620.
